Enzymatic fragmentation and peptide spectrometry through nanopores (ProNanoPep)

The goal of the project is to develop a new approach that combines highly controlled and automated enzymatic fragmentation with a new principle, peptide spectrometry through nanopores, for label-free identification, discrimination and ultimately sequencing of proteins.

The goal is to explore a novel platform for label-free protein identification, discrimination and ultimately sequencing. The platform consists of:

  1. A microfluidic approach for automated enzymatic fragmentation of polypeptides.
  2. A biohybrid sensor consisting of a protein nanopore inserted into a biomimetic phospholipid membrane. An array of such nanopore devices is provided directly on a CMOS-based, low-noise current amplifier array.
