Platform for automated high sensitive protein analytics for the quantification of drug agents and biomarkers in the field of drug development (dINA)

The goal of project dINA is the development of a platform for droplet based high sensitive protein analytics for the quantification of drug agents and biomarkers to be used in the field of drug development. As part of this a centrifugal microfluidic chip will be designed and manufactured using injection molding at Hahn-Schickard.

The goal of project dINA is the development of a platform for droplet based high sensitive protein analytics for the quantification of drug agents and biomarkers to be used in the field of drug development. Precise quantification of drug agents and biomarkers as well as the detection of substances leading to unspecific immune responses is very important in the field of drug development. As a standard method for quantification ELISA (enzyme-linked immunosorbent assay) is commonly used. Using this technology it can be a challenge to reach high levels of analytical sensitivity as well as a high dynamic range.
Instead of using ELISA, the goal in the project dINA is the quantification of proteins by a newly developed digital Immuno- loop-mediated isothermal amplification (LAMP) (Chimera Biotec GmbH, Mast Diagnostica GmbH). This allows a precise quantification with high sensitivity and high dynamic range across 105 magnitudes.

For digitalization and automation of the immono-LAMP assay a centrifugal-microfluidic chip for the generation of thousands of reaction compartments (droplets) will be designed and manufactured using injection molding at Hahn-Schickard. To complete the platform an automated processing device will be developed by the partner Biofluidix GmbH.
